Inspiration is a game rule that allows a player to infuse a little bit of luck into their game.
The DM can give Inspiration to characters if they feel the player has played the character as realistically as possible.
Characters have a range of attributes (background, religion, traits - even spells and skills) that make them unique - and also make them as real as possible. It is up to the player to use these attributes as best they can.
For example, a cleric that stops at a local temple dedicated to his god and leaves a tithe is doing what their character would do in a real word situation - and that deserves some Inspiration!
To use the Inspiration, a player can choose to roll a second d20 on any attack roll, saving throw, or ability check - then apply the advantage rules to the dice.
Once used, the Inspiration is gone - And a character either does or does not have Inspiration - It doesn't accrue.
A player can also choose to gift their Inspiration to another player if they feel they deserve it. This has to be done during rest (not during a battle!)
More information on the Inspiration can be found on page 125 of the 5e Player's Handbook.
